Ramanna joined the Tata Institute of Fundamental Research (TIFR) in Mumbai, where he worked under the guidance of renowned physicist Homi J. Bhabha. He was involved in India's first peaceful nuclear explosion, codenamed "Smiling Buddha," which took place on May 18, 1974, at the Pokhran Test Range in Rajasthan. This event made India a nuclear weapons state.

Raja Ramanna served as the Director of the Bhabha Atomic Research Centre (BARC) in Mumbai from 1972 to 1978.
